Tryptic digestion (Tryptic Digest)

Tryptic digestion is a widely used process for splitting proteins into smaller peptides. It uses the enzyme trypsin, which specifically cuts the peptide bonds after the amino acids lysine and arginine. This method is particularly useful for breaking down large proteins into manageable peptide fragments, which can then be further analysed. It plays a central role in proteomics, the study of proteins and their functions (bottom-up method).

 

After tryptic digestion, the resulting peptide fragments are often analysed using high-performance liquid chromatography (HPLC). HPLC makes it possible to separate the different peptides based on their size, polarity or charge. By combining HPLC with other analytical methods, such as mass spectrometry (HPLC-MS), proteins can be characterised in detail. This is important for the identification of proteins, the investigation of post-translational modifications and for quantitative analyses in biological samples.
