Buffer
Buffers are indispensable components in chromatography that stabilise the pH value in the mobile phase. This is particularly important as the pH value significantly influences the retention properties of the analytes and the separation performance of the chromatographic column. The use of buffers can significantly improve the reproducibility and accuracy of chromatographic analyses.
Technical Data
In chromatography, especially in high-performance liquid chromatography (HPLC), various buffers are used to ensure optimum separation conditions. Frequently used buffers include
Phosphate buffers: phosphate buffers are very popular due to their wide pH range (approx. 2 to 12) and buffering capacity. They are often used in reversed-phase HPLC.
Acetate buffers: These buffers are particularly suitable for the separation of analytes at a pH value between 3.5 and 5.5. They are often used in ion exchange chromatography.
Tris buffer: Tris(hydroxymethyl)aminomethane (Tris) is a good buffer for the pH range from 7 to 9 and is often used in biochemical applications.
Formate buffer: Formate buffers are useful for pH values in the range of 2.8 to 4.5 and are used in the separation of organic acids.
