In chromatography, especially in high-performance liquid chromatography (HPLC), various buffers are used to ensure optimum separation conditions. Frequently used buffers include

 

Phosphate buffers: phosphate buffers are very popular due to their wide pH range (approx. 2 to 12) and buffering capacity. They are often used in reversed-phase HPLC.

 

Acetate buffers: These buffers are particularly suitable for the separation of analytes at a pH value between 3.5 and 5.5. They are often used in ion exchange chromatography.

 

Tris buffer: Tris(hydroxymethyl)aminomethane (Tris) is a good buffer for the pH range from 7 to 9 and is often used in biochemical applications.

 

Formate buffer: Formate buffers are useful for pH values in the range of 2.8 to 4.5 and are used in the separation of organic acids.
