OligoMg

OligoMg columns from Sielc have been specially developed for the analysis of oligonucleotides. These columns are also based on the Bridge Ion Separation Technique (BIST) and have a negatively charged surface loaded with magnesium 2+ (Mg2+). In the presence of an organic modifier, these cations on the stationary phase enable the interaction between the negatively charged oligonucleotide and the stationary phase. The magnesium cations act as a bridge. This has the advantage that oligonucleotides can be effectively separated with a mobile phase consisting only of water and methanol.

Influence of the water content on the retention of oligonucleotides with the OligoMg from Sielc

Based on BIST technology, the OligoMg column has dual positively charged ions loaded onto the surface. This means that it is no longer necessary to add these divalent ions to the mobile phase. This means that oligonucleotides can be separated without the addition of a buffer

Separation of different 6mer oligonucleotides with the OligoMG of Sielc

The geometry of different oligonucleotides influences the bridge binding between analytes and the stationary phase and thus the retention. In addition to the geometry, the strength and composition of the nucleobases also play a decisive role. This allows different oligonucleotides with the same chain length to be separated from each other.
