Monolithic Columns
Unlike other columns,monolithic columns do not consist of individual particles. Instead, they consist of a single rod of highly porous material. This porous material has macropores and mesopores. The macropores allow the mobile phase to flow through the material at low back pressure (pore size >1 µm). The mesopores are narrower and enable high-performance separation (pore size 100-300 Å). Monolithic columns are available as monoliths based on silica and on organic polymers.
What are the advantages of monolithic columns?
- Low back pressure at high flow rates - shortens the analysis time
- Column efficiency does not decrease significantly at higher flow rates
- Columns last longer than fully porous particle columns due to low back pressure
- Monolithic columns are very robust
- High matrix tolerance - no complex sample preparation required

With the Chromolith series, Merck offers monolithic silica-based columns. They are available in different pore sizes (macro- and mesopores) and various modifications. The dimensions range from capillary columns to preparative dimensions.
2. Thermo Scientific
Thermo Scientific offers monolithic columns based on organic polymers for various bio-applications. The ProSwift and PepSwift series are available in various modifications and dimensions.
